چکیده
Mantle cell lymphoma (MCL) is a difficult-to-treat B-cell malignancy characterized by cyclin D1 (CD1) overexpression. Targeting CD1 in MCL has been shown to be of therapeutic significance. However, treatment remains challenging since patients are still subject early and frequent relapse the disease. To ensure their high proliferation rate, tumour cells have increased iron needs, making them more susceptible deprivation. Indeed, several chelators proved effective anti-cancer agents. In this study, we demonstrate that clinically approved chelator deferasirox (DFX) exerts an anti-tumoural effect lines patient cells. The exposure feasible concentrations DFX resulted growth inhibition, cycle arrest induction apoptosis. We show unfolds its cytotoxic rapid reactive oxygen species (ROS) leads oxidative stress severe DNA damage triggering proteolysis mechanism requires phosphorylation on T286 glycogen synthase kinase-3? (GSK3?). Moreover, mediates repressing phosphatidylinositol 3-kinase (PI3K)/AKT/GSK3? pathway via ROS generation. Our data suggest as potential option for paves way options these patients.

Targeting cyclin D1 for mantle cell lymphoma
Cyclin D1, an important component of cell cycle and a protein with known oncogenic potential, is over expressed in mantle cell lymphoma (MCL). MCL is a distinct clinical pathologic subtype of B cell non-Hodgkin’s lymphoma often associated with poor prognosis. New therapeutic approaches based on boosting anti-tumor immunity are being developed. Targeting cyclin D1 for MCL is rendering an interes...

In this issue of Blood, Salaverria et al report that more than half of Cyclin D1- (CCND1) negative SOX11-positive mantle cell lymphoma (MCL) had CCND2 gene rearrangement predominantly with immunoglobulin (IG) light chain genes.(1)

Objective: Cuscuta campestris or common dodder is a holoparasitic plant that has been valorized for treatment of liver injury and cancer prevention in traditional medicine. Recently, extract of C. campestris had shown moderate antimicrobial properties and cytotoxic effects. In this study, we examined the level of cellular oxidants, cytotoxicity, apoptosis and differentiation induced by hydr...
متن کاملCyclin D1-negative mantle cell lymphoma with cryptic t(12;14)(p13;q32) and cyclin D2 overexpression.
Virtually all cases of mantle cell lymphoma (MCL) carry the t(11;14)(q13;q32) translocation, leading to the juxtaposition of the CCND1/CYCLIND1 gene to the immunoglobulin heavy chain (IGH) joining region, resulting in cyclin D1 mRNA and protein overexpression.1-3 The existence of “true” MCL negative for cyclin D1 has been controversial but was recently substantiated by gene expression profiling...

process of activation, division, and differentiation to generate a large pool of activated effector T cells. Regulated induction of apoptosis of these expanded T cells avoids several problems for the host, including increased metabolic cost, disruption of lymphoid homeostasis, and predisposition to autoimmunity and lymphoid neoplasia. On the other hand, it is essential that some activated T cel...
